Harnessing Open-Source Solutions: Insights From the First Open Systems Pharmacology (OSP) Community Conference

JournalPBPK/PBBMPharmacometricsQSP

About the publication

This publication summarize sessions from the first Open OSP Community Conference (Basel, Switzerland, October 7–8, 2024). The conference brought together scientists, modelers, and industry professionals to share developments and practical applications in this evolving field.

Key topics:

  • Translation of in vitro measures into clinical pharmacokinetics using PBPK and PBBM
  • PBPK modeling for special populations, particularly in obstetric populations
  • A comprehensive, quantitative systems pharmacology (QSP) model for predicting subcutaneous absorption, bioavailability, and immunogenicity
  • Regulatory requirements and interactions, supported by case examples of how OSP has been applied in regulatory contexts

Co-authored by: Pharmetheus’ Marylore Chenel, Tobias Kanacher and Erik Sjögren, together with Paul Vrenken, André Dallmann at Bayer, and Ibrahim Ince at Boehringer Ingelheim.

About OSP

Open Systems Pharmacology (OSP) is an open-source, community-driven initiative focused on advancing PBPK and QSP modeling. Best known for tools like PK-Sim and MoBi, the platform supports model-informed drug development and is widely used in both research and regulatory settings.

Pharmetheus has long maintained a close and active relationship with the OSP community, contributing to the development and practical application of its tools. As a consultancy focused on model-informed drug development, Pharmetheus regularly integrates OSP software — like PK-Sim® and MoBi® — into its pharmacokinetic modeling and simulation projects. Several members of the Pharmetheus team are involved in the OSP community. Notably, Erik Sjögren, who is also affiliated with Uppsala University, serves on the OSP management team, helping to guide the platform’s strategic direction and future growth.

In addition to its technical contributions, Pharmetheus is actively engaged in the broader OSP ecosystem through hands-on workshops and training initiatives, and shares a strong commitment to advancing open-source modeling tools and promoting their use in both industry and regulatory settings.
