Physiologically based pharmacokinetic (PBPK) models and quantitative systems pharmacology (QSP) models are built on prior knowledge, including (patho-)physiology, and pharmacology. The independent representations of drug and system specific parameters allow for extrapolation to scenarios beyond observation.
